Descripción

FNIP2, a homolog of FNIP1, functions as a secondary but critical binding partner of folliculin (FCLN) in metabolic regulation and energy sensing. Like FNIP1, FNIP2 contributes to the stability of the FCLN-FNIP complex and facilitates the proper localization and activation of mTORC1 signaling in response to nutrient availability. FNIP2 has overlapping roles with FNIP1, including interactions with AMPK and Hsp90, highlighting its importance in maintaining cellular energy balance. Its expression patterns are similar to FNIP1, being prominent in tissues with high metabolic demand, such as skeletal muscle, kidney, and liver. Studies suggest that FNIP2 may have redundant or compensatory functions with FNIP1, as loss of FNIP2 alone has less severe phenotypic consequences compared to combined FNIP1/FNIP2 deficiencies. FNIP2's role in immune cell metabolism and its involvement in B-cell differentiation suggest potential implications in immune regulation. Despite functional overlap, FNIP2 exhibits unique roles in specific tissues and under stress conditions, emphasizing its distinct but complementary contributions to folliculin-mediated pathways.
