Descripción

DENN domain containing 2A (DENND2A) is a guanine nucleotide exchange factor (GEF) that activates Rab9, a key regulator of vesicular transport between late endosomes and the trans-Golgi network. The DENN domain in DENND2A facilitates the exchange of GDP for GTP on Rab9, promoting vesicle tethering, sorting, and fusion events critical for retrograde transport. DENND2A plays an essential role in maintaining lysosomal and endosomal integrity by ensuring the proper recycling of receptors and transport of lysosomal hydrolases. This protein is vital for the degradation and recycling of membrane proteins, contributing to cellular signaling and nutrient uptake. Dysregulation of DENND2A has been implicated in lysosomal storage disorders and diseases involving defective vesicular trafficking, as its dysfunction can disrupt the balance of cargo transport and endosomal recycling. Its broad expression across tissues, combined with its role in maintaining vesicular trafficking, highlights its importance in cellular homeostasis and adaptive responses to environmental signals.
