Descripción

DOCK4 is a GEF that activates Rac1 and Rap1, regulating cell adhesion, migration, and junction formation. It is critical for maintaining epithelial integrity and angiogenesis by promoting actin cytoskeletal remodeling. DOCK4 is widely expressed in tissues, including the brain, and has roles in neuronal development, synapse formation, and dendritic growth. Mutations in DOCK4 are linked to cancers, particularly prostate, ovarian, and colorectal cancers, as they disrupt cell adhesion and enhance invasion. It interacts with proteins like ELMO1 for Rac1 activation and mediates processes such as cell spreading. Knockdown studies show impaired junction stability and migration, emphasizing its role in tissue organization and metastasis.
