Ameloblastin, encoded by the AMBN gene, is a protein primarily found in the extracellular matrix of dental tissues, particularly in enamel-forming cells called ameloblasts. It is a major component of the enamel matrix, which is essential for the formation and mineralization of dental enamel, the outer layer of teeth. Ameloblastin plays several important roles in tooth development and enamel formation. Matrix organization: Ameloblastin helps organize the enamel matrix, providing a scaffold for the deposition of mineral crystals during enamel formation. It interacts with other enamel matrix proteins and forms complexes that contribute to the structural integrity of the developing enamel. Mineralization regulation: Ameloblastin participates in the regulation of enamel mineralization by influencing the nucleation and growth of hydroxyapatite crystals, the main mineral component of enamel. It contains specific domains that can bind calcium and phosphate ions, which are essential for the mineralization process. Cell signaling: Ameloblastin can also act as a signaling molecule, influencing the behavior and function of ameloblasts during enamel development. It interacts with cell surface receptors and intracellular signaling pathways, modulating processes such as cell adhesion, proliferation, and differentiation.
