Background

AKR1A1 is a widely expressed enzyme that belongs to the aldo-keto reductase superfamily. It catalyzes the reduction of aldehydes and ketones to their respective alcohols using NADPH as a cofactor, playing a key role in detoxifying reactive aldehydes generated during lipid peroxidation and metabolism. AKR1A1 is involved in the metabolism of endogenous compounds, such as prostaglandins, steroids, and lipid-derived aldehydes, as well as exogenous toxins, including chemotherapeutics and environmental pollutants. AKR1A1 has significant antioxidant properties due to its ability to reduce harmful aldehydes, thus protecting cells from oxidative stress. It also contributes to glucose metabolism by participating in the sorbitol pathway, particularly in the liver, kidneys, and lens. Dysregulation of AKR1A1 is implicated in metabolic disorders, including diabetic complications, cataract formation, and cardiovascular diseases. Additionally, AKR1A1 has been shown to play a role in drug resistance by metabolizing chemotherapeutic agents, making it a target for improving cancer treatments.
