AKAP12, also known as gravin, is a multifunctional scaffold protein that binds PKA, PKC, and other kinases, organizing signaling pathways at the plasma membrane and cytoskeleton. AKAP12 plays a critical role in cell adhesion, migration, and proliferation by coordinating cAMP and PKC signaling in focal adhesions. It regulates cytoskeletal remodeling and cell polarity by interacting with actin-binding proteins, influencing processes like wound healing and cancer metastasis. AKAP12 acts as a tumor suppressor in various cancers, where its loss leads to dysregulated signaling pathways, including ERK/MAPK and cell cycle progression. AKAP12 is also involved in vascular homeostasis, where it regulates endothelial barrier function and angiogenesis. Studies highlight AKAP12’s role in stress responses, particularly in regulating PKA signaling during inflammation and oxidative stress. Additionally, AKAP12 expression is regulated by epigenetic mechanisms, such as promoter methylation, which contributes to its silencing in cancer cells.
