Background

AK8, localized in the cytoplasm and nuclear compartments, is a nucleotide kinase involved in nucleotide homeostasis and cellular energy regulation. AK8 specifically catalyzes the phosphotransfer reaction between ATP/GTP and AMP to sustain ATP regeneration. Its nuclear localization highlights a unique role in chromatin remodeling, DNA repair, and RNA synthesis processes, where energy supply is tightly regulated. AK8 expression is particularly enriched in rapidly dividing cells, such as immune cells, stem cells, and certain cancer cells, supporting proliferation and metabolic reprogramming. Dysregulation of AK8 has been associated with tumorigenesis due to its role in maintaining nucleotide pools for DNA replication. In neurons, AK8 also regulates energy homeostasis, influencing processes such as neurotransmitter synthesis and synaptic plasticity.
