Description
Initiates extracellular glutathione (GSH) breakdown, provides cells with a local cysteine supply and contributes to maintain intracelular GSH level. It is part of the cell antioxidant defense mechanism. Catalyzes the transfer of the glutamyl moiety of glutathione to amino acids and dipeptide acceptors. Alternatively, glutathione can be hydrolyzed to give Cys-Gly and gamma glutamate.

Gamma-Glutamyltransferase 1 (GGT1) is a membrane-bound enzyme that catalyzes the transfer of gamma-glutamyl moieties from glutathione to amino acids and peptides, playing a critical role in glutathione homeostasis and antioxidant defense GGT1 is essential for the breakdown and recycling of glutathione, a key molecule in detoxifying reactive oxygen species (ROS) and maintaining cellular redox balance GGT1 is highly expressed in the kidney, liver, and other epithelial tissues, where it regulates amino acid transport and cellular responses to oxidative stress Dysregulation of GGT1 activity is associated with oxidative damage, inflammation, and metabolic diseases, including liver disorders, cardiovascular disease, and diabetes Elevated GGT1 levels are also observed in cancers such as hepatocellular carcinoma and lung cancer, where they promote tumor progression and chemoresistance through enhanced glutathione metabolism Additionally, GGT1 serves as a biomarker for liver function and oxidative stress-related conditions Its role in glutathione metabolism and cellular redox regulation underscores its importance in maintaining cellular health and defense mechanisms
