Fc Receptor Like 3 (FCRL3) is expressed on B cells, regulatory T cells, and some natural killer cells, functioning as an immune checkpoint molecule. The presence of ITIMs in its structure supports its inhibitory role in immune regulation, particularly in dampening autoimmunity. FCRL3 polymorphisms have been linked to autoimmune diseases such as rheumatoid arthritis and type 1 diabetes, where genetic variants contribute to increased susceptibility. Collectively, the FCRL family represents a critical set of regulators for immune cell function, offering potential therapeutic targets for autoimmune disorders and B cell-related malignancies.
