DPH7 functions as a regulatory factor that ensures the fidelity and efficiency of the diphthamide biosynthetic process. While not directly involved in enzymatic reactions, DPH7 aids in the recognition and stabilization of intermediates, particularly during the amidination step catalyzed by DPH6. Structural and functional analyses suggest that DPH7 interacts with eEF2 and other biosynthetic enzymes, maintaining the integrity of the diphthamide synthesis complex. Loss of DPH7 function disrupts the maturation of diphthamide, rendering cells resistant to diphtheria toxin and reducing overall protein synthesis efficiency. DPH7 is broadly expressed, with high levels in tissues with active protein translation and energy metabolism. Emerging studies propose additional roles for DPH7 in cellular stress responses and protein quality control pathways. Mutations in DPH7 are associated with subtle defects in cellular growth and homeostasis, underscoring its auxiliary but critical role in the biosynthesis of diphthamide.
