Descripción
DENN domain containing 2C (DENND2C) acts as a guanine nucleotide exchange factor (GEF) for Rab GTPases, regulating intracellular vesicle trafficking and membrane dynamics. DENND2C activates Rab9 by facilitating the exchange of GDP for GTP, enabling vesicular transport between late endosomes and the trans-Golgi network. This activity is essential for the sorting and trafficking of lysosomal enzymes and other cargo, maintaining cellular proteostasis. In addition to its role in endosomal transport, DENND2C contributes to signaling pathways that depend on membrane trafficking, including those involved in cell growth and stress responses. DENND2C is expressed in various tissues, but its activity is particularly significant in cells with high lysosomal turnover, such as macrophages and epithelial cells. Dysregulation of DENND2C has been linked to diseases involving lysosomal dysfunction, highlighting its importance in cellular homeostasis and adaptive responses.
