Description
The protein encoded by this gene is one of the signaling and transforming proteins containing Src homology 2 and 3 (SH2 and SH3) domains. It is located in the cytoplasm and is an adaptor protein involved in transducing signals from receptor tyrosine kinases to downstream signal recipients such as RAS. Alternatively spliced transcript variants encoding different isoforms have been found.

Background
NCK1 is a cytoplasmic adaptor protein containing three SH3 domains and one SH2 domain, which allows it to mediate interactions between receptor tyrosine kinases (RTKs) and downstream effectors in signaling pathways. NCK1 is involved in actin cytoskeleton reorganization, cell motility, and immune synapse formation by linking signaling proteins such as WASP, WAVE, and PAK1. It plays key roles in processes like cell migration, proliferation, and T-cell activation. NCK1 is highly expressed in various tissues and regulates growth factor receptor signaling, including pathways like MAPK/ERK and PI3K. Dysregulation of NCK1 is associated with cancer progression, metastasis, and immune system dysfunction. Knockout studies show defects in actin dynamics, impaired cell movement, and reduced T-cell activation, highlighting its importance in cytoskeletal signaling and cellular communication.
