BCL2 interacting protein 3 (BNIP3) is a pro-apoptotic member of the Bcl-2 family that plays a key role in regulating cell death, particularly in response to cellular stress BNIP3 functions by interacting with anti-apoptotic Bcl-2 family members, promoting mitochondrial outer membrane permeabilization, and facilitating the release of pro-apoptotic factors such as cytochrome c from the mitochondria This release activates the caspase cascade, triggering apoptosis BNIP3 is particularly involved in regulating hypoxia-induced cell death and autophagy, which are essential for cellular adaptation to low oxygen levels and nutrient stress Dysregulation of BNIP3 has been linked to various diseases, including cancer and neurodegenerative disorders, where altered expression of BNIP3 contributes to uncontrolled cell survival, tumor progression, and impaired cellular homeostasis BNIP3's role in mitochondrial function and its involvement in the hypoxic response make it an important factor in regulating cell death and adaptation to environmental stresses
