Non-essential component of the volume-regulated anion channel(VRAC, also named VSOAC channel), an anion channel required to maintain a constant cell volume in response to extracellular or intracellular osmotic changes. The VRAC channel conducts iodide better than chloride and may also conduct organic osmolytes like taurine. Channel activity requires LRRC8A plus at least one other family member(LRRC8B, LRRC8C, LRRC8D or LRRC8E); channel characteristics depend on the precise subunit composition(PubMed:24790029, PubMed:26824658). LRRC8A and LRRC8D are required for the uptake of the drug cisplatin(PubMed:26530471). Mediates the import of the antibiotic blasticidin-S into the cell(PubMed:24782309).


Background

LRRC8D is one of the five subunits of the volume-regulated anion channel (VRAC), which plays a crucial role in cellular osmoregulation and volume control. This channel complex enables the efflux of chloride ions and organic osmolytes in response to cell swelling, maintaining cellular integrity under osmotic stress. Structurally, LRRC8D contains leucine-rich repeats (LRRs) in its extracellular domain, which contribute to its ability to interact with other VRAC subunits, including LRRC8A, the essential core component. LRRC8D is known for its unique substrate specificity, particularly its ability to facilitate the transport of certain drugs, such as cisplatin, and organic molecules like taurine and amino acids. Dysregulation of LRRC8D function has been implicated in chemoresistance, highlighting its role in cancer therapeutics. Furthermore, LRRC8D is crucial in the regulation of extracellular signaling and apoptosis, particularly in cells experiencing stress-induced swelling. Its importance extends to physiological processes such as kidney function, where osmolyte regulation is critical, making it a target of interest in pharmacological studies aiming to modulate VRAC activity.
