Description
The nuclear import of karyophilic proteins is directed by short amino acid sequences termed nuclear localization signals (NLSs). Karyopherins, or importins, are cytoplasmic proteins that recognize NLSs and dock NLS-containing proteins to the nuclear pore complex. The protein encoded by this gene shares the sequence similarity with Xenopus importin-alpha and Saccharomyces cerevisiae Srp1. This protein is found to interact with the NLSs of DNA helicase Q1 and SV40 T antigen.

Descripción
KPNA4, also known as importin-α3, is a nuclear transport adaptor protein that recognizes classical nuclear localization signals (NLS) in cargo proteins and facilitates their nuclear import via the importin-β pathway. KPNA4 binds cargo proteins, such as transcription factors and kinases, forming a complex that translocates through the nuclear pore complex (NPC). It is widely expressed and plays critical roles in cell cycle regulation, transcriptional control, and stress responses. KPNA4 is implicated in viral replication by mediating the nuclear import of viral proteins in pathogens like HIV and influenza virus. Dysregulation of KPNA4 has been linked to cancers, where it enhances nuclear transport of oncogenic proteins and supports tumor progression. Knockdown studies show its role in maintaining proper cellular signaling, highlighting its importance in development, differentiation, and immune responses.
