Description

The Mediagnost anti-HAV IgM ELISA is a "class-capture" enzyme immuoassay. Serum or plasma samples are diluted 1:2000 and added to the wells of a microtiter plate, which have been previously coated with an antibody directed against human IgM antibodies (µ-chain specific). All IgM antibodies present in the sample are bound during an incubation step. After washing, the Hepatitis A-virus is added, which binds only to the HAV-specific IgM antibodies. The bound antigen is detected by the addition of the conjugate (monoclonal anti-HAV antibody, peroxidase conjugated). After washing and incubation with a colorimetric substrate the reaction is terminated by addition of stop solution and the color turns to yellow. The absorbance of the coloured reaction product is measured on a microtiter plate reader. The colour intensity of the reaction corresponds to the concentration of antibodies in the sample

Mediagnost anti-VHA IgM Elisa es un inmunoensayo enzimático de "captura ". Las muestras de suero o plasma se diluyen 1:2000 y se añaden a los pocillos de una placa de microtitulación, previamente recubierta con un anticuerpo dirigido contra anticuerpos IgM humanos (específicos de la cadena µ). Todos los anticuerpos IgM presentes en la muestra se unen durante una etapa de incubación. Tras el lavado, se añade el virus de la hepatitis A, que se une únicamente a los anticuerpos IgM específicos del VHA. El antígeno unido se detecta mediante la adición del conjugado (anticuerpo monoclonal anti-VHA conjugado con peroxidasa). Tras el lavado y la incubación con un sustrato colorimétrico, la reacción se detiene mediante la adición de solución de parada y el color se torna amarillo. La absorbancia del producto de reacción coloreado se mide en un lector de placas de microtitulación. La intensidad del color de la reacción corresponde a la concentración de anticuerpos en la muestra.

The detection of anti-HAV IgM (IgM antibodies specific for Hepatitis A virus) in the serum or plasma of patients indicates a fresh infection with the Hepatitis-A virus. Depending on the antiHAV IgM titre it is possible to differentiate between an acute infection (1 - 3 months after the start of clinical symptoms) and early convalescence (3 - 6 months). In rare cases (approx. 10% of all clinical cases) the anti-HAV IgM response remains positive (6 - 12 months).
