Description

DOK proteins are enzymatically inert adaptor or scaffolding proteins. They provide a docking platform for the assembly of multimolecular signaling complexes. DOK5 functions in RET-mediated neurite outgrowth and plays a positive role in activation of the MAP kinase pathway. Putative link with downstream effectors of RET in neuronal differentiation.

Descripción

DOK5 is a member of the DOK family, functioning as an adaptor protein in signaling pathways downstream of RET and insulin receptors. It is expressed in skeletal muscle, heart, and nervous tissue, where it regulates processes like cell survival, differentiation, and metabolic signaling. DOK5 enhances insulin signaling by activating the MAPK/ERK pathway but does not influence the PI3K/Akt pathway. It is critical for neurite outgrowth and neuronal development, particularly in response to neurotrophic factors. Dysregulation of DOK5 contributes to insulin resistance and is implicated in neurological disorders, where it affects neuronal survival and axonal guidance.
