Description

DOK proteins are enzymatically inert adaptor or scaffolding proteins. They provide a docking platform for the assembly of multimolecular signaling complexes. DOK3 is a negative regulator of JNK signaling in B-cells through interaction with INPP5D/SHIP1. May modulate ABL1 function(By similarity).

Descripción

Docking protein 3 (DOK3) is a unique member of the DOK family, characterized by its selective expression in lymphoid tissues and its critical role in B-cell receptor (BCR) signaling. Unlike DOK1 and DOK2, DOK3 predominantly functions in modulating adaptive immune responses. It acts as a negative regulator of BCR signaling by interacting with various kinases and phosphatases, thereby preventing hyperactivation of B cells. This function is crucial for maintaining immune tolerance and preventing autoimmune disorders. DOK3 has also been implicated in the regulation of inflammation and the production of cytokines in macrophages. Dysregulation of DOK3 expression or function has been linked to immune deficiencies and certain lymphoid malignancies, highlighting its importance in immune regulation and cellular signaling pathways.
