Description
Acyl-CoA thioesterases are a group of enzymes that catalyze the hydrolysis of acyl-CoAs to the free fatty acid and coenzyme A(CoASH), providing the potential to regulate intracellular levels of acyl-CoAs, free fatty acids and CoASH. Displays high levels of activity on medium-and long chain acyl CoAs.

Descripción
ACOT2 is a mitochondrial enzyme that catalyzes the hydrolysis of acyl-CoA esters into free fatty acids and CoA, playing a pivotal role in mitochondrial fatty acid metabolism and energy regulation. It is highly expressed in energy-demanding tissues such as the heart, liver, and skeletal muscle, where it regulates the pool of acyl-CoA intermediates and ensures efficient β-oxidation. ACOT2 also serves as a key regulator of mitochondrial CoA homeostasis, preventing the accumulation of acyl-CoA esters that could inhibit mitochondrial enzymes. Its role extends to regulating lipid signaling molecules and providing precursors for membrane biosynthesis. Dysregulation of ACOT2 has been associated with metabolic dysfunctions, such as insulin resistance and mitochondrial disorders, reflecting its importance in maintaining energy balance and metabolic integrity. ACOT2’s mitochondrial localization and activity highlight its critical role in coordinating lipid utilization and energy production under various physiological conditions.
