Description

Component of the shelterin complex(telosome) that is involved in the regulation of telomere length and protection. Shelterin associates with arrays of double-stranded TTAGGG repeats added by telomerase and protects chromosome ends. Without its protective activity, telomeres are no longer hidden from the DNA damage surveillance and chromosome ends are inappropriately processed by DNA repair pathways. Promotes binding of POT1 to single-stranded telomeric DNA. Modulates the inhibitory effects of POT1 on telomere elongation. The ACD-POT1 heterodimer enhances telomere elongation by recruiting telomerase to telomeres and increasing its processivity. May play a role in organogenesis.

Descripción

ACD, also known as TPP1, is a shelterin complex component involved in protecting telomeres and maintaining chromosomal stability. It regulates telomere length and integrity by recruiting telomerase and interacting with other shelterin proteins, such as POT1, to prevent inappropriate DNA damage responses at chromosome ends. ACD plays a critical role in cellular aging, genome stability, and cancer suppression. It is expressed in proliferative tissues, where telomere maintenance is essential for cellular replication. Dysregulation or mutations in ACD are associated with telomere-related disorders, such as dyskeratosis congenita and aplastic anemia, as well as increased susceptibility to cancer due to compromised chromosomal stability. ACD’s functions in telomere biology underscore its importance in aging, genome integrity, and disease prevention.
