Background
ANKRD16 is a protein containing multiple ankyrin repeat domains, which mediate protein-protein interactions and play key roles in cellular processes such as transcriptional regulation and protein stability. ANKRD16 has been identified as a tRNA splicing factor that interacts with components of the tRNA splicing machinery to ensure proper RNA maturation and protein synthesis. Loss of ANKRD16 function disrupts tRNA homeostasis, leading to the accumulation of defective RNA species and cellular stress. ANKRD16 is also involved in maintaining proteostasis and preventing the accumulation of misfolded proteins, which has been linked to neurodegenerative disorders. Recent studies suggest its role as a neuroprotective factor due to its ability to regulate tRNA quality control pathways, and mutations or dysregulation of ANKRD16 have been associated with neurodevelopmental diseases and certain cancers.
