Descripción
ANAPC11 is the catalytic RING finger subunit of the Anaphase-Promoting Complex/Cyclosome (APC/C), where it facilitates the ubiquitin transfer activity of the complex. As an E3 ubiquitin ligase, ANAPC11 directly interacts with E2 ubiquitin-conjugating enzymes to catalyze the ubiquitination of cell cycle regulators such as cyclin B and securin, promoting their degradation via the proteasome. This process is critical for initiating anaphase, regulating mitotic exit, and maintaining genomic stability. ANAPC11 is essential for APC/C activity, and its loss or inhibition results in mitotic arrest, aneuploidy, and cell cycle defects. Mutations or dysregulation of ANAPC11 have been associated with cancer progression, as impaired proteolysis of APC/C substrates can lead to uncontrolled proliferation and chromosomal instability. ANAPC11’s catalytic function makes it a central component of APC/C and a potential target for cancer therapies aimed at disrupting mitotic progression in tumor cells.
