Background

ALDH8A1 is a cytosolic enzyme primarily involved in the metabolism of retinaldehyde to retinoic acid, a key signaling molecule that regulates cell differentiation, growth, and tissue homeostasis. ALDH8A1 is expressed in metabolic tissues, including the liver, kidneys, and retina, where it supports retinoid metabolism and vitamin A homeostasis. Its role in converting retinaldehyde to retinoic acid makes ALDH8A1 critical for maintaining vision, embryonic development, and immune responses. Dysregulation of ALDH8A1 has been implicated in retinal diseases, such as age-related macular degeneration (AMD), and developmental abnormalities caused by retinoic acid deficiency. ALDH8A1 also participates in detoxifying aldehydes derived from lipid peroxidation and xenobiotic metabolism, contributing to cellular defense against oxidative stress. Emerging studies suggest its involvement in maintaining retinoid signaling pathways and redox balance.
