Descripción
ALDH1L2 is the mitochondrial isoform of 10-formyltetrahydrofolate dehydrogenase and plays a critical role in mitochondrial one-carbon metabolism. It catalyzes the oxidation of 10-formyltetrahydrofolate to CO2 and tetrahydrofolate, producing NADPH, which supports mitochondrial redox balance and biosynthetic processes. ALDH1L2 is essential for maintaining nucleotide synthesis, amino acid metabolism, and methylation pathways within the mitochondria, particularly during high energy and biosynthetic demands. Its function in producing NADPH also makes it critical for detoxifying reactive oxygen species (ROS) and protecting cells from oxidative stress. Dysregulation of ALDH1L2 has been implicated in metabolic diseases, neurodegenerative conditions, and cancer. In cancer, loss of ALDH1L2 contributes to metabolic reprogramming, where mitochondrial folate metabolism is disrupted to favor rapid proliferation and tumor growth. Recent studies indicate that ALDH1L2 could serve as a therapeutic target for rebalancing mitochondrial metabolism and reducing oxidative stress in cancer cells.
