AK6, also known as GTP:AMP phosphotransferase, is a mitochondrial matrix enzyme primarily responsible for transferring phosphate groups between nucleotide molecules, such as GTP and AMP. AK6 plays a critical role in mitochondrial nucleotide homeostasis, facilitating ADP and ATP regeneration to sustain oxidative phosphorylation. Unlike cytosolic AK isoforms, AK6's GTP preference highlights its importance in maintaining mitochondrial energy pools. It is essential for cellular processes dependent on mitochondrial energy, including protein synthesis, RNA processing, and DNA replication.
