Description
ACOX1 is the first enzyme of the fatty acid beta-oxidation pathway, which catalyzes the desaturation of acyl-CoAs to 2-trans-enoyl-CoAs. It donates electrons directly to molecular oxygen, thereby producing hydrogen peroxide. Defects in this gene result in pseudoneonatal adrenoleukodystrophy, a disease that is characterized by accumulation of very long chain fatty acids.

Acyl-CoA oxidases (ACOXs) are crucial enzymes involved in the first step of peroxisomal β-oxidation, a pathway responsible for the breakdown of very-long-chain fatty acids (VLCFAs), branched-chain fatty acids, and bile acid intermediates. ACOXs catalyze the oxidation of acyl-CoA to trans-2-enoyl-CoA, producing hydrogen peroxide in the process. ACOX catalyzes the first step of the beta-oxidation pathway within peroxisomes. It oxidizes long-chain acyl-CoA molecules, removing two carbons from the fatty acid chain to produce trans-2-enoyl-CoA. This reaction involves the transfer of electrons to molecular oxygen, leading to the formation of hydrogen peroxide (H2O2). Fatty acid beta-oxidation is a crucial pathway for the catabolism of fatty acids, which are important sources of energy for cells. By breaking down fatty acids into acetyl-CoA units, beta-oxidation provides substrates for energy production through processes such as the citric acid cycle and oxidative phosphorylation. ACOX1 preferentially oxidizes straight-chain VLCFAs.
