AKAP5 Antibody is a Rabbit Polyclonal antibody against AKAP5. The A-kinase anchor proteins (AKAPs) are a group of structurally diverse proteins, which have the common function of binding to the regulatory subunit of protein kinase A (PKA) and confining the holoenzyme to discrete locations within the cell. This gene encodes a member of the AKAP family. The encoded protein binds to the RII-beta regulatory subunit of PKA, and also to protein kinase C and the phosphatase calcineurin. It is predominantly expressed in cerebral cortex and may anchor the PKA protein at postsynaptic densities (PSD) and be involved in the regulation of postsynaptic events. It is also expressed in T lymphocytes and may function to inhibit interleukin-2 transcription by disrupting calcineurin-dependent dephosphorylation of NFAT.


Background

AKAP5, also known as AKAP79/150, is a cytosolic scaffold protein that anchors PKA, PKC, and calcineurin to the plasma membrane in neurons, cardiac tissue, and epithelial cells. AKAP5 is highly enriched in synapses, where it organizes signaling complexes that regulate synaptic plasticity, neurotransmitter release, and calcium signaling. It interacts with AMPA and NMDA receptors, ensuring the spatial regulation of PKA and PKC signaling necessary for synaptic strength and long-term potentiation (LTP), processes critical for learning and memory. AKAP5 also plays a role in cardiac function, where it regulates ion channels, such as L-type calcium channels, influencing cardiac excitation-contraction coupling. Dysregulation of AKAP5 disrupts neuronal signaling, leading to cognitive deficits, psychiatric disorders, and neurodegenerative diseases. Additionally, AKAP5 regulates receptor internalization and trafficking, highlighting its role in cellular signaling and membrane dynamics.
