Multiple receptor subtypes of serotonin neurotransmitters with multiple physiologic functions have been recognized. The 5-HT-2 receptors mediate many of the central and peripheral physiologic functions of serotonin. Cardiovascular effects include contraction of blood vessels and shape changes in platelets; central nervous system effects include neuronal sensitization to tactile stimuli and mediation of hallucinogenic effects of phenylisopropylamine hallucinogens.


Background

HTR2B is another subtype of the G protein-coupled serotonin receptors that modulates diverse physiological functions, including cardiovascular function, smooth muscle contraction, and mood regulation. HTR2B is expressed in the central nervous system (CNS) and peripheral tissues, particularly in the heart, liver, and gastrointestinal tract. In the CNS, it plays a role in mood stabilization and reward processing by regulating the release of neurotransmitters like dopamine. HTR2B is also critical for the development and maintenance of cardiac function, as its activation leads to cardiac hypertrophy and structural remodeling. Dysregulation of HTR2B has been linked to pulmonary hypertension, where its overactivation promotes abnormal vasoconstriction and vascular remodeling. It is also implicated in drug-induced valvular heart disease, particularly from medications like fenfluramine and MDMA (ecstasy), which act as HTR2B agonists. HTR2B’s interaction with serotonergic pathways suggests its relevance in mood disorders, impulsivity, and addiction. Genetic variants in HTR2B have been associated with behavioral abnormalities, such as aggressive impulsivity. Although less studied compared to HTR2A, HTR2B is increasingly recognized as a therapeutic target for cardiovascular disorders, mood stabilization, and conditions involving abnormal serotonin signaling, such as valvular diseases and pulmonary vascular conditions.
