Descripción

OXGR1, also known as the G protein-coupled receptor 99 (GPR99), is activated by 2-oxoglutarate (α-ketoglutarate), an important intermediate in the Krebs cycle. It is predominantly expressed in the kidney, lungs, and heart, where it regulates acid-base homeostasis, metabolic sensing, and inflammatory responses. OXGR1 couples to Gq proteins, activating phospholipase C (PLC), increasing intracellular calcium levels, and modulating downstream signaling pathways. In the kidney, OXGR1 plays a key role in the regulation of bicarbonate transport and pH balance, particularly in response to metabolic acidosis. OXGR1 has also been implicated in inflammation, as α-ketoglutarate regulates immune responses through its signaling in inflammatory cells. Dysregulation of OXGR1 is associated with metabolic disorders, kidney dysfunction, and inflammatory diseases. Its emerging role as a sensor for metabolic intermediates positions OXGR1 as a potential therapeutic target for metabolic acidosis, kidney diseases, and immune-related disorders.
