PolyclonalDescripción
YWHAB belongs to the 14-3-3 family of proteins, members of which mediate signal transduction by binding to phosphoserine-containing proteins. This highly conserved protein family is found in both plants and mammals. The encoded protein has been shown to interact with RAF1 and CDC25 phosphatases, suggesting that it may play a role in linking mitogenic signaling and the cell cycle machinery. Two transcript variants differing in the 5' UTR, but encoding the same protein, have been identified for the gene. Both variants encode the same protein, however, they are differentially expressed in hematopoietic cells.
Background
YWHAB, part of the 14-3-3 protein family, functions as a scaffold protein that binds to phosphorylated serine/threonine-containing motifs, regulating protein localization, stability, and signaling pathways. It is involved in processes such as cell cycle control, DNA damage response, apoptosis, and stress adaptation by interacting with key signaling proteins, including Raf kinases, BAD, and CDC25. YWHAB regulates pathways like MAPK and PI3K/Akt, influencing cellular growth, survival, and metabolism. It is ubiquitously expressed, with high levels in the brain, where it contributes to synaptic plasticity, neuroprotection, and neuronal development. Dysregulation of YWHAB has been associated with neurodegenerative diseases, including Alzheimer’s and Parkinson’s, as well as cancer, where altered protein interactions promote tumor survival and metastasis. Knockout studies show defects in cellular proliferation, increased apoptosis, and impaired stress responses, highlighting its role in maintaining cellular stability and signaling balance.
