Apolipoprotein E (APOE) is a protein encoded by the APOE gene located on chromosome 19 in humans. APOE is a polymorphic protein, meaning it can exist in different isoforms encoded by three common alleles: ε2, ε3, and ε4. These isoforms differ in their amino acid sequences, resulting in variations in protein structure and function. The primary function of APOE is to facilitate the uptake and clearance of lipoproteins, such as chylomicrons, very low-density lipoproteins (VLDL), and remnants of these lipoproteins, from the bloodstream. APOE acts as a ligand for cell surface receptors, including the low-density lipoprotein receptor (LDLR) and the LDL receptor-related protein (LRP), mediating the uptake of lipoprotein particles into cells. In addition to its role in lipid metabolism, APOE is involved in various physiological processes, including neuronal repair, synaptic plasticity, and inflammation modulation. In the central nervous system, APOE is synthesized by astrocytes and microglia and is critical for the maintenance and repair of neuronal membranes. It plays a role in the clearance of amyloid beta peptides, which are implicated in the pathogenesis of Alzheimer's disease.The expression of the APOE gene can be influenced by various factors, including dietary nutrients, hormones, and transcriptional regulators.   • Secondary Structure: The protein predominantly consists of alpha-helices.
  • Tertiary Structure: APOE has a globular structure with a distinct N-terminal domain responsible for receptor binding and a C-terminal domain involved in lipid binding.
  • Quaternary Structure: APOE functions as a monomer but can form dimers or multimers, particularly when lipid-bound.


Classification and Subtypes:

APOE belongs to the apolipoprotein family, which includes various proteins that bind lipids to form lipoproteins. APOE exists in three major isoforms, which differ by single amino acid substitutions:

  • APOE2 (Cys112, Cys158)
  • APOE3 (Cys112, Arg158)
  • APOE4 (Arg112, Arg158)

These isoforms have different biochemical properties and associations with disease risk.
