Biological Activity: A549 cells were seeded into 96 well plates (4,000 cells/well) with DMEM + 5% serum and different concentrations of recombinant TNFA. After incubation for 72 h, cells were observed by inverted microscope and cell proliferation was measured using a Cell Counting Kit-8 (CCK-8). 10 μl of CCK-8 solution was added to each well of the plate, then the absorbance at 450 nm was measured using a microplate reader after incubating the plate for 1h at 37°C. The EC50 was dermined to be 3.02 µg/ml, and TNFA was shown to significantly increase the viability of A549 cells.

To keep the original salt concentration, we recommend reconstituting to the original concentration prior to lyophilization (see Concentration) in ddH2O. If a lower concentration is required, dilute in 10 mM PBS, pH 7.4. If a higher concentration is required, the product can be reconstituted directly in 10 mM PBS, pH 7.4, though please note that this will change the overall salt concentration. The stock concentration should be between 0.1-1.0 mg/ml. Do not vortex.
