UBX domain protein 2B (UBXN2B) is a paralog of UBXN2A that also contains a UBX domain, enabling interaction with p97/VCP for the regulation of protein degradation pathways UBXN2B primarily participates in the extraction and clearance of misfolded proteins through endoplasmic reticulum-associated degradation (ERAD) and proteasomal pathways It plays a role in maintaining proteostasis, particularly under conditions of cellular stress, where protein turnover and quality control mechanisms are critical UBXN2B has been shown to influence apoptotic pathways by regulating p97-dependent protein degradation, ensuring appropriate cellular responses to stress Dysregulation of UBXN2B contributes to neurodegenerative diseases where defective protein clearance leads to protein aggregation and cellular toxicity Recent studies also indicate a role for UBXN2B in cancer biology, where it affects tumor cell survival and growth by modulating protein homeostasis and stress responses Its ability to regulate proteasome function highlights its importance in maintaining cellular integrity during proteotoxic stress and disease conditions
