Background
UBX domain protein 11 (UBXN11) is a member of the UBX family of proteins that interact with p97/VCP, a central ATPase in protein degradation and cellular quality control pathways The UBX domain in UBXN11 enables its binding to p97, facilitating the regulation of processes like endoplasmic reticulum-associated degradation (ERAD), protein turnover, and ubiquitin-proteasome pathways UBXN11 also plays a role in autophagy by modulating protein extraction and lysosomal targeting during cellular stress Its involvement in maintaining proteostasis ensures the removal of misfolded or aggregated proteins, critical for cell survival Dysregulation of UBXN11 is linked to neurodegenerative diseases such as Alzheimer’s and Huntington’s, where defective protein clearance contributes to disease progression UBXN11 is also implicated in cancer biology, where its role in protein turnover and stress response can influence tumor growth, survival, and therapy resistance Emerging evidence highlights its potential function in cellular signaling pathways, particularly those associated with proteotoxic stress, autophagy, and DNA damage repair
