Descripción

SOCS6 is a member of the SOCS protein family that regulates cytokine and growth factor signaling pathways, particularly those mediated by insulin and receptor tyrosine kinases (RTKs). It inhibits signaling by targeting receptor complexes and signaling intermediates for ubiquitin-mediated degradation via its SOCS box domain, thus fine-tuning pathways such as the PI3K/Akt and MAPK cascades. SOCS6 is broadly expressed in tissues like the liver, pancreas, and skeletal muscle, where it regulates processes like cell growth, glucose metabolism, and insulin sensitivity. It has a role in tumor suppression by inhibiting growth factor signaling and promoting apoptosis in cancer cells. Dysregulation of SOCS6 is linked to metabolic disorders, including insulin resistance and diabetes, as well as cancer progression due to impaired signaling control. Knockdown studies reveal enhanced RTK activity, increased cellular proliferation, and reduced apoptosis, highlighting its role as a regulator of metabolic signaling, growth, and tumor suppression.

The protein encoded by this gene contains a SH2 domain and a CIS homolog domain. The protein thus belongs to the cytokine-induced STAT inhibitor (CIS), also known as suppressor of cytokine signaling (SOCS) or STAT-induced STAT inhibitor (SSI), protein family. CIS family members are known to be cytokine-inducible negative regulators of cytokine signaling. The expression of this gene can be induced by GM-CSF and EPO in hematopoietic cells. A high expression level of this gene was found in factor-independent chronic myelogenous leukemia (CML) and erythroleukemia (HEL) cell lines.
