Descripción
SOCS5 acts as a negative regulator of cytokine signaling by inhibiting JAK/STAT pathways and targeting signaling components for proteasomal degradation through its SOCS box domain. It interacts with cytokine receptors like IL-4Rα to regulate T-helper cell differentiation, particularly inhibiting Th2 responses and promoting immune balance. SOCS5 is expressed in lymphoid tissues, epithelial cells, and adipose tissues, where it modulates immune regulation, cytokine sensitivity, and energy homeostasis. It is particularly important for regulating allergic inflammation and T-cell polarization during immune responses. Dysregulation of SOCS5 is associated with allergic diseases, asthma, and autoimmune disorders due to imbalanced cytokine signaling. Functional studies reveal its role in controlling IL-4-driven responses, preventing excessive Th2-mediated inflammation. Knockout models demonstrate enhanced Th2 differentiation, increased allergic inflammation, and disrupted immune balance, emphasizing its role in maintaining cytokine homeostasis and adaptive immunity.
