Descripción

SOCS4 is a member of the SOCS protein family that regulates cytokine signaling by inhibiting the Janus kinase (JAK)/STAT pathway and promoting the proteasomal degradation of signaling intermediates via its SOCS box domain. It interacts with phosphorylated cytokine receptors and downstream proteins to suppress excessive cytokine responses, particularly in pathways related to epidermal growth factor receptor (EGFR) and interleukin-4 (IL-4). SOCS4 is broadly expressed in tissues such as the liver, immune cells, and epithelial tissues, where it regulates processes like cell proliferation, differentiation, and immune homeostasis. It has been implicated in regulating inflammation and epithelial repair during infections. Dysregulation of SOCS4 is linked to chronic inflammation, impaired wound healing, and cancer progression, where excessive EGFR signaling promotes uncontrolled cell proliferation. Knockdown studies reveal hyperactivated cytokine signaling, increased inflammatory responses, and abnormal cellular growth, highlighting its critical role in balancing cytokine-mediated signaling pathways.

The protein thus belongs to the suppressor of cytokine signaling (SOCS), also known as STAT-induced STAT inhibitor (SSI), protein family. SOCS family members are known to be cytokine-inducible negative regulators of cytokine signaling. Two alternatively spliced transcript variants encoding the same protein have been found for this gene.
