Signal-transducing adaptor protein 2 (STAP2) is an adaptor protein that plays a significant role in intracellular signaling pathways by linking various receptors to downstream effectors, particularly within immune and inflammatory responses. It contains a PH domain, SH2 domain, and a proline-rich region, which facilitate its interaction with tyrosine-phosphorylated proteins and its localization to membrane-associated signaling complexes. STAP2 is implicated in the regulation of cytokine signaling pathways, such as those mediated by STAT3 and STAT5, influencing processes like cell survival, proliferation, and differentiation. It has been shown to interact with components of the T-cell receptor complex and Fc receptors, modulating immune responses and contributing to signal transduction in lymphocytes and macrophages. Dysregulation of STAP2 expression has been linked to pathological conditions, including autoimmune diseases, chronic inflammation, and cancer, where it may promote oncogenic signaling or alter immune surveillance.
