Descripción

SH3 Domain Containing Kinase Binding Protein 1 (SH3KBP1), also known as CIN85 (Cbl-interacting protein of 85 kDa), is a multi-domain adaptor protein involved in numerous cellular signaling pathways. It contains SH3 domains that facilitate interactions with various binding partners, making it a critical regulator of receptor endocytosis, cytoskeletal reorganization, and intracellular trafficking. SH3KBP1 plays a key role in downregulating activated receptor tyrosine kinases, such as EGFR, by recruiting ubiquitin ligases like Cbl, leading to receptor internalization and degradation. Additionally, it is implicated in actin cytoskeleton dynamics, thereby influencing processes such as cell migration and adhesion. SH3KBP1 also participates in immune signaling and synaptic transmission by modulating vesicular transport. Dysregulation of SH3KBP1 has been associated with cancer, particularly in pathways involving aberrant receptor signaling, as well as other diseases linked to defective endocytic or cytoskeletal mechanisms.
