Inhibitory serpins have been shown to function in processes as diverse as DNA binding, chromatin condensation, immunoregulation ,apoptosis and haemostasis. Serpins are a broadly distributed protease inhibitors that use the conformational change as a mechanism of action. They are central in controlling many important proteolytic cascades. Phylogenetically Serpins superfamily organized in "Clades", named by letters. The following numbers shows the position in the glade. Serpin named as a SERPINXy, (where X is the clade and y is the number within that clade) many serpins also have alternative names from before this classification was proposed. Structuraly Serpinsare three β-sheets (A, B and C), 8-9 α-helices (termed hA-hI) and the reactive center known as reactive center loop (RCL.). SERPINA12, a 47 kDa protease inhibitor from the adipokine family, has insulin-sensitizing properties. Its treatment in obese and insulin-resistant mice reduces insulin resistance gene expression in white adipose tissue and improves carbohydrate resistance.
