SAM and SH3 domain containing 1 (SASH1) is a scaffolding protein that functions as a tumor suppressor and a signaling adaptor in cellular processes such as adhesion, migration, and proliferation SASH1 contains SAM (Sterile Alpha Motif) and SH3 (Src Homology 3) domains that enable interactions with signaling molecules, linking it to pathways regulating cytoskeletal organization, inflammation, and apoptosis It is predominantly expressed in epithelial tissues and immune cells, where it modulates cell-cell adhesion and response to extracellular signals Loss or downregulation of SASH1 expression is associated with tumor progression, invasion, and poor prognosis in cancers such as breast, colorectal, and lung cancers, underscoring its role in suppressing metastasis SASH1 is also involved in regulating inflammatory pathways and immune responses, suggesting its broader role in maintaining tissue homeostasis Emerging studies highlight its importance in vascular remodeling and wound healing, where it influences endothelial cell function Its ability to interact with multiple signaling pathways makes SASH1 a key regulator of cell behavior and a potential therapeutic target in cancer and inflammatory diseases
