Background

JADE2 is a member of the JADE family of PHD finger proteins, functioning as a cofactor in chromatin remodeling and histone acetylation. It is a component of the HBO1 (histone acetyltransferase binding to ORC1) complex, where it regulates H4 acetylation, particularly during DNA replication initiation and cell cycle progression. JADE2 binds to acetylated histones and modulates transcriptional activation, chromatin accessibility, and cellular proliferation. It is expressed in various tissues, including actively dividing cells, where it regulates gene expression linked to cell cycle progression and DNA replication. Dysregulation of JADE2 is associated with genomic instability and tumorigenesis, as altered histone acetylation disrupts chromatin dynamics and transcriptional control. Knockdown studies reveal defects in histone acetylation, impaired cell cycle progression, and disrupted DNA replication initiation, highlighting its role in epigenetic regulation and cellular proliferation.
