Recombinant Human DCAF1

Background

DDB1 and CUL4 associated factor 1 (DCAF1), also known as VprBP, is a multifunctional protein that serves as a substrate receptor within the CUL4-DDB1 E3 ubiquitin ligase complex. It plays a pivotal role in various cellular processes, including cell cycle regulation, telomerase activity, and histone modification. DCAF1's interaction with the HIV-1 accessory protein Vpr has been extensively studied, revealing its involvement in viral pathogenesis. Structurally, DCAF1 contains WD40 repeats that facilitate protein-protein interactions, enabling it to recruit specific substrates for ubiquitination. Its expression is ubiquitous, with notable presence in the nucleoplasm and nucleoli fibrillar center, indicating its involvement in nuclear functions. DCAF1's kinase activity, particularly its ability to phosphorylate histone H2A at threonine 120, underscores its role in transcriptional regulation. Additionally, DCAF1 is implicated in the regulation of microRNA biogenesis and the development and function of B and T lymphocytes, highlighting its significance in both general and cell type-specific processes.
