PPARGC1A, also known as PGC-1α (Peroxisome proliferator-activated receptor gamma coactivator 1-alpha), is a transcriptional coactivator that plays a central role in the regulation of cellular energy metabolism and mitochondrial biogenesis. PPARGC1A interacts with and coactivates various transcription factors, including peroxisome proliferator-activated receptor gamma (PPARG) and nuclear respiratory factors (NRFs), to regulate the expression of genes involved in energy metabolism, oxidative phosphorylation, and mitochondrial function. It promotes the transcription of genes involved in fatty acid oxidation, glucose metabolism, and thermogenesis, thereby enhancing cellular energy production and expenditure. PPARGC1A is a master regulator of mitochondrial biogenesis, the process by which new mitochondria are formed. It stimulates the expression of nuclear-encoded mitochondrial genes and activates signaling pathways involved in mitochondrial biogenesis, such as the PPAR signaling pathway and the AMP-activated protein kinase (AMPK) pathway. Increased PPARGC1A activity leads to the proliferation and remodeling of mitochondria, resulting in enhanced cellular energy capacity and metabolic efficiency.
