Descripción
NMUR2 is a G protein-coupled receptor activated by neuromedin U (NMU) and is predominantly expressed in the central nervous system (CNS), particularly in the hypothalamus, brainstem, and spinal cord. NMUR2 couples to Gq/11 proteins, activating downstream pathways such as phospholipase C (PLC) and calcium signaling. NMUR2 plays a critical role in regulating energy balance, feeding behavior, and stress responses. In the hypothalamus, NMUR2 activation suppresses appetite and enhances energy expenditure, linking it to the regulation of obesity and metabolic homeostasis. NMUR2 is also involved in the modulation of pain perception and stress-induced behaviors, with emerging evidence suggesting its role in anxiety and inflammatory pain pathways. Dysregulation of NMUR2 has been associated with metabolic disorders, stress-related diseases, and chronic pain conditions. NMUR2 agonists are being explored as potential therapeutic agents for obesity and metabolic syndrome, while antagonists may be beneficial for pain and stress-related disorders, highlighting its dual role in energy regulation and pain modulation.
