Nuclear receptor subfamily 3 group C member 2 (NR3C2), also known as the mineralocorticoid receptor (MR), is a nuclear receptor that mediates the effects of mineralocorticoids, particularly aldosterone, which regulates salt and water balance, blood pressure, and electrolyte homeostasis NR3C2 is primarily expressed in the kidneys, heart, and brain, where it controls the reabsorption of sodium and water, influencing blood volume and pressure It functions by binding to aldosterone and translocating to the nucleus, where it regulates the expression of genes involved in electrolyte balance and fluid retention NR3C2 is essential for maintaining blood pressure and regulating the body's response to dehydration and salt imbalances Dysregulation of NR3C2 has been linked to various cardiovascular and renal disorders, including hypertension, heart failure, and kidney disease Overactivation of MR signaling can lead to fluid retention and hypertension, while decreased activity can result in conditions like hypoaldosteronism In addition, MR is a target for drugs such as mineralocorticoid antagonists, which are used to treat conditions like heart failure and hypertension

Mutations in this gene cause autosomal dominant pseudohypoaldosteronism type I, a disorder characterized by urinary salt wasting. Defects in this gene are also associated with early onset hypertension with severe exacerbation in pregnancy. Alternative splicing results in multiple transcript variants.
