Descripción
LRRC7, also known as Densin-180, is a postsynaptic scaffolding protein enriched in the brain, where it regulates synaptic organization, signaling, and plasticity. It interacts with key proteins such as PSD-95, CaMKII, and actin regulators to stabilize and organize the postsynaptic density (PSD) at excitatory synapses. LRRC7 plays an essential role in dendritic spine maturation, synaptic strength, and calcium signaling, contributing to processes like learning, memory, and neuronal development. It is particularly abundant in regions like the hippocampus and cortex, where synaptic plasticity is critical. Dysregulation or mutations in LRRC7 are associated with neurodevelopmental disorders, including intellectual disabilities and autism spectrum disorders (ASD), due to impaired synaptic connectivity and function. Knockout studies in mice reveal altered dendritic spine morphology, impaired synaptic transmission, and deficits in learning and memory, highlighting its essential role in maintaining synaptic structure and cognitive function.
