Descripción
P2RY4 is a G protein-coupled receptor activated by extracellular UTP and ATP. It predominantly couples to Gq proteins, stimulating phospholipase C (PLC) and increasing intracellular calcium levels, leading to activation of downstream signaling pathways. P2RY4 is expressed in epithelial tissues, including the gastrointestinal tract, kidney, and respiratory tract, where it regulates ion transport, fluid secretion, and epithelial cell function. In the intestine, P2RY4 plays a role in chloride and fluid secretion, contributing to mucosal hydration and barrier function. It is also involved in inflammation and tissue repair, particularly in response to extracellular nucleotides released during tissue damage. Dysregulation of P2RY4 signaling has been linked to gastrointestinal disorders, cystic fibrosis, and epithelial dysfunction. Targeting P2RY4 with selective modulators holds therapeutic potential for treating intestinal inflammation, respiratory diseases, and conditions involving impaired ion transport and mucosal hydration.
