Protein Tyrosine Kinase 2 (PTK2), also known as FAK (Focal Adhesion Kinase), is a non-receptor tyrosine kinase that plays a critical role in cell adhesion, migration, and signal transduction. PTK2 is primarily located at focal adhesion sites and is activated in response to integrin signaling and growth factor receptors. Structurally, PTK2 consists of an N-terminal FERM domain for membrane association, a central kinase domain for catalytic activity, and a C-terminal focal adhesion targeting (FAT) domain that mediates protein-protein interactions. PTK2 is crucial in cytoskeletal organization, integrin-mediated signaling, and cell motility, particularly in processes such as embryogenesis, wound healing, and angiogenesis. PTK2 is upregulated in various cancers, including breast, colorectal, and glioblastoma, where it promotes cell proliferation, survival, and metastasis. Dysregulation of PTK2 signaling can contribute to chemoresistance and increased tumor progression, making it a potential therapeutic target. In non-cancer contexts, PTK2 is essential for immune cell function, particularly for T-cell activation and macrophage responses. Additionally, PTK2 is implicated in neurological processes, such as synaptic plasticity and neuron survival.
