Prostaglandin D2 Synthase (PTGDS) is an enzyme that catalyzes the isomerization of prostaglandin H2 (PGH2) to prostaglandin D2 (PGD2), a key lipid mediator involved in sleep regulation, allergic inflammation, vasodilation, and immune modulation PTGDS is highly expressed in the brain, heart, and male reproductive organs, where it participates in cerebrospinal fluid production, detoxification of free radicals, and maintenance of male fertility PGD2 plays a central role in allergic reactions, mediating bronchoconstriction in asthma and inflammation in rhinitis Dysregulation of PTGDS contributes to neurological disorders such as Alzheimer’s disease by facilitating tau phosphorylation, amyloid deposition, and oxidative stress PTGDS also serves as a biomarker for cardiovascular disease, brain injury, and central nervous system inflammation Its role in lipid signaling and antioxidant defense highlights its importance in both homeostasis and disease progression

The protein encoded by this gene is a glutathione-independent prostaglandin D synthase that catalyzes the conversion of prostaglandin H2 (PGH2) to postaglandin D2 (PGD2). PGD2 functions as a neuromodulator as well as a trophic factor in the central nervous system. PGD2 is also involved in smooth muscle contraction/relaxation and is a potent inhibitor of platelet aggregation. This gene is preferentially expressed in brain. Studies with transgenic mice overexpressing this gene suggest that this gene may be also involved in the regulation of non-rapid eye movement sleep.
